本論文之研究主要針對自臺灣海域採集之隔板葉形軟珊瑚Lobophytum crassum及皮軟珊瑚Briareum stechei兩種八放珊瑚所含天然物成分進行分析,其中從隔板葉形軟珊瑚L. crassum中分離純化出2個cembranoid類的雙萜類化合物,包含一個新化合物lobocrassin I (1) 與1個已知化合物lobohedleolide (2) 。另外,從皮軟珊瑚B. stechei中分離純化出3個briarane-type類的雙萜類化合物,包含1個已知化合物excavatolide M (3) 與兩個新化合物briastecholide K (4) 、briastecholide L (5)。
化合物1–5的結構皆透過核磁共振光譜資料分析與文獻資料比對方式加以解析建立,此外化合物1、2和3更進一步經由X-ray單晶體繞射實驗來證明其化合物之絕對立體結構。
化合物2針對脂多醣 (lipopolysaccharide, LPS) 所誘發小鼠巨噬細胞RAW264.7產生一氧化氮合成酶 (inducible nitric oxide synthase, iNOS) 之發炎性蛋白具有抑制活性。根據西方墨點法實驗結果顯示,化合物2在10 μM濃度下能抑制iNOS,使其蛋白表現量下降至28.50%。
In the continuing studies on the chemical constituents of octocorals distributed in the waters of Formosan, two cembranoids, including a new compound, lobocrassin I (1), along with a known analogue, lobohedleolide (2) were isolated from an octocoral identified as Lobophytum crassum. In addition, a know briarane-type diterpenoid, excavatolide M (3), as well as two new briaranes, briastecholides K (4) and L (5), were obtained from Briareum stechei.
Spectroscopic approaches were used to reveal the structures of compounds 1–5, and the structures, including absolute configurations of 1–3 were further confirmed by single-crystal X-ray diffraction analysis.
Cembranoid 2 exhibited activity against the protein expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-treated RAW264.7 mouse macrophage cells. Western blotting results showed that 2 at 10 μM inhibited iNOS expression, which reduced the protein level to 28.50%.
